چکیده

Background: Alcohol misuse represents a substantial risk to insurance companies. The Centers for Disease Control (CDC) identities alcohol as the second (to tobacco) most prevalent avoidable public health risk with alcoholic cirrhosis and hepatitis being the most common liver diseases seen in clinical practice.1 Public Health reported prevalence of chronic alcohol misuse in the general population is 2.8%, (range 1.2 to 5.6%).2 CDC surveys report that the prevalence of alcohol misuse is negatively correlated with age, education, and income. The reported prevalence of alcohol misuse in the affluent (higher than median income), older (older than 45 years), and educated population (graduated from college) is less than 2%.3

Harmonization of measurement results of the alcohol biomarker carbohydrate-deficient transferrin by use of the toolbox of technical procedures of the International Consortium for Harmonization of Clinical Laboratory Results.

BACKGROUND The need for equivalent results of routine measurement procedures for the alcohol biomarker carbohydrate-deficient transferrin (CDT) has been recognized by the IFCC. This article describes a project to harmonize CDT as conducted by an IFCC working group initiated for this purpose. METHODS We used procedures for achieving harmonization as developed by the Consortium for Harmonizatio...

Toward standardization of carbohydrate-deficient transferrin (CDT) measurements: II. Performance of a laboratory network running the HPLC candidate reference measurement procedure and evaluation of a candidate reference material.

Carbohydrate-deficient transferrin (CDT) is a descriptive term used for a temporary change in the transferrin glycosylation profile caused by alcohol, and used as a biomarker of chronic high alcohol consumption. The use of an array of methods for measurement of CDT in various absolute or relative amounts, and sometimes covering different transferrin glycoforms, has complicated the comparability...

Determination of carbohydrate-deficient transferrin in human serum by capillary zone electrophoresis: evaluation of assay performance and quality assurance over a 10-year period in the routine arena.

The performance of high-resolution CZE for determination of carbohydrate-deficient transferrin (CDT) in human serum based on internal and external quality data gathered over a 10-year period is reported. The assay comprises mixing of serum with a Fe(III) ion-containing solution prior to analysis of the iron saturated mixture in a dynamically double-coated capillary using a commercial buffer at ...

Carbohydrate-deficient transferrin measured by high-performance liquid chromatography and CDTect immunoassay.

Carbohydrate-deficient transferrin (CDT) was measured in three populations using anion-exchange chromatography followed by radioimmunoassay (CDTectTM) and a new high-performance liquid chromatography (HPLC) method. The correlation between the methods in 50 consecutive clinical samples was good (r = 0.87). However, in a set of 49 samples with CDT concentrations close to the reference value of th...

Validation by isoelectric focusing of the anion-exchange isotransferrin fractionation step involved in determination of carbohydrate-deficient transferrin by the CDTect assay.

Serum concentration of carbohydrate-deficient transferrin (CDT) is used for laboratory diagnosis of chronic alcohol abuse. Using isoelectric focusing for validation of the initial isotransferrin fractionation step involved in the determination of CDT by the CDTect assay, we found a complete in vitro iron saturation of transferrin and sufficient stability of the transferrin iron load during colu...

Development and multicenter evaluation of the N latex CDT direct immunonephelometric assay for serum carbohydrate-deficient transferrin.

BACKGROUND Carbohydrate-deficient transferrin (CDT) is a promising biomarker of alcohol abuse. We describe the development and multicenter evaluation of N Latex CDT (Dade Behring), an automated, particle-enhanced, homogeneous immunonephelometric assay for directly determining CDT. METHODS N Latex CDT uses a monoclonal antibody that recognizes the structure of transferrin glycoforms lacking 1 ...
